Abstract

Black rice bran is a food ingredient with unique health benefits. It contains many active compounds, particularly anthocyanins, natural substances that help protect the body from damage. This study aimed to optimize the extraction process parameters, including microwave power, extraction time, and solvent volume, using the Microwave Assisted Extraction (MAE) method to achieve optimal extract yield, anthocyanin content, color, and pH. The experiment was designed using Response Surface Methodology (RSM) with a Box-Behnken Design. The three independent variables were microwave power (300–600 Watts), extraction time (5–15 minutes), and solvent volume (600–1500 mL), while the observed responses included extract yield, anthocyanin content, color value (L), and pH. The optimization results showed that the best conditions were obtained at 423.436 Watts, 12 minutes, and 600 mL solvent volume, resulting in a yield of 7.95%, anthocyanin content of 416 ppm, an L value of 8.25, and pH of 4.62. Moderate increases in power and extraction time improved the release of anthocyanins, while excessive solvent volume decreased anthocyanin concentration and extract color intensity. The low pH indicates the acidic condition that helps maintain anthocyanin stability. Therefore, MAE proved to be an effective and efficient method for extracting anthocyanins from black rice bran in a relatively short time.
