Abstract

The purpose of this study was to determine as the effect of the concentration of nitrogen source addition of urea and adding pure concentration of the yeast Saccharomyces cerevisiae tape. The research method consists of two phases: a preliminary study and the main study. The preliminary study was conducted to determine the optimum growth of Saccharomyces cerevisiae in YPG liquid medium (yeast peptone glucose) and urea concentration of 2.5%, 5% and 7.5%. The primary research consisted of, physical analysis that measures the pH value, chemical analysis, namely the reduction sugar, microbiological analysis that calculates the number of living cells with a counting chamber. The design of experiment is used is a randomized block design (RAK) with factorial pattern of 3x3 as much as 3 repetitions, followed further test of Duncan for real different factors. Factors trial consists of urea concentration ratio of 2.5%, 2%, 1% and the concentration of Saccharomyces cerevisiae 0.5 ml, 0.75 ml, 1 ml. Results showed that the preliminary study obtained optimum growth time on the clock 39 17.4 x 106 cells / ml and the addition of a nitrogen source the best that urea concentration of 2.5%. Interaction concentration of Saccharomyces cerevisiae and urea concentration effect on growth of yeast Saccharomyces cerevisiae in the manufacture of tape.
